Thus, targeting lectins, especially those on the surface of immune cells, could advance immunology and drug discovery. Lectins are typically oligomeric; therefore, many of the most potent ligands are multivalent. An effective strategy for lectin targeting is to display multiple copies of a single glycan epitope on a polymer backbone; however, a drawback to such multivalent ligands is they cannot distinguish between lectins that share monosaccharide binding selectivity (e.g., mannose-binding lectins such as DC-SIGN, DC-SIGNR, MBL, SP-D, langerin, dectin-2, mincle, and DEC-205) as they often lack molecular precision.<\/p>\n<p>Authors developed \u03b2-mannosylated glyco-IEGmers specific to each lectins with mannose binding specificity.<\/p>\n<p>To generate the target mannosylated glycomacromolecules, iterative exponential growth (IEG) cycles beginning from (R)- or (S)-glycidyl propargyl ether (GPE, > 99% ee) were conducted to yield oligo\/polytriazoles with precisely 8, 16, or 32 allyl side chains. Macromolecules were produced with three different overall absolute configurations: all (R) (\u201cisotactic\u201d), all (S) (\u201cisotactic\u201d), and alternating (R-alt-S) (\u201csyndiotactic\u201d). To append the mannose residues, these IEGmers were exposed to pure \u03b2-thiomannose sodium salt under UV light (\u03bb = 365 nm).<\/p>\n<p><a href=\"https:\/\/www.emukk.com\/WP\/wp-content\/uploads\/2021\/11\/mannosylated-glyco-IGEmers\"><img loading=\"lazy\" decoding=\"async\" src=\"https:\/\/www.emukk.com\/WP\/wp-content\/uploads\/2021\/11\/mannosylated-glyco-IGEmers-300x110.png\" alt=\"\" width=\"300\" height=\"110\" class=\"alignnone size-medium wp-image-6718\" srcset=\"https:\/\/www.emukk.com\/WP\/wp-content\/uploads\/2021\/11\/mannosylated-glyco-IGEmers-300x110.png 300w, https:\/\/www.emukk.com\/WP\/wp-content\/uploads\/2021\/11\/mannosylated-glyco-IGEmers-600x220.png 600w, https:\/\/www.emukk.com\/WP\/wp-content\/uploads\/2021\/11\/mannosylated-glyco-IGEmers-1024x375.png 1024w, https:\/\/www.emukk.com\/WP\/wp-content\/uploads\/2021\/11\/mannosylated-glyco-IGEmers-768x281.png 768w, https:\/\/www.emukk.com\/WP\/wp-content\/uploads\/2021\/11\/mannosylated-glyco-IGEmers.png 1048w\" sizes=\"auto, (max-width: 300px) 100vw, 300px\" \/><\/a><\/p>\n<p>The binding affinity changed from sample to sample over several orders of magnitudes. For example, K<sub>A<\/sub> values for DC-SIGN ranged from approximately 1 \u00d7 10<sup>4<\/sup> for (S)-8mer to about 1 \u00d7 10<sup>8<\/sup> for (R)-16mer. For dectin-2, it also ranged from \u223c1 \u00d7 10<sup>4<\/sup> to \u223c1 \u00d7 10<sup>8<\/sup>. DEC-205 bound all of the glyco-IEGmers with high K<sub>A<\/sub> values in the range of \u223c1 \u00d7 10<sup>6<\/sup> to 1 \u00d7 10<sup>9<\/sup>, with the highest K<sub>A<\/sub> of all pairwise interactions tested (2.2 \u00d7 10<sup>9<\/sup>). <\/p>\n<p>This wide range in affinity is impressive as the mannose binding epitopes presented was identical among these
